EFFECT OF ANNONA MURICATA LINNFRUIT JUICE ON RENAL ISCHEMIA/REPERFUSION-INDUCED BIOCHEMICAL DYSFUNCTION IN HYPERLIPIDEMIC RATS

dc.description.abstract This study was designed to investigatethe effect of Annona muricata Linn fruit juice on renal ischemia/reperfusion-induced biochemical dysfunction in hyperlipidemic rat.Annona muricata fruits was cut into smaller pieces and then pulverized with a blender. For in vivo experiment, 60 male Wistar rats were divided into 10 groups (group 2-5 was administered Triton while group 1 served as control, group 7-10 was administered triton plus renal ischemia reperfusion while group 6 served as sham) rats were intoxicated with 200 mg/kg Triton and post-treated with 200 mg/kg and 400 mg/kg of Annona muricata fruit juice (AMFJ) daily, for 14 days and renal ischemia was carried out for the ischemia groups on the 13th day followed by biochemical estimations in serum and tissues (heart and kidney) as well as histopathological evaluation of the kidney.The result of the in vivo study showed that AMFJ significantly ameliorated the alterations in serum lipid profiles occasioned by Triton intoxication as adjudged by its effects on total cholesterol, triglycerides, high density lipoprotein-cholesterol, low density lipoprotein-cholesterol, and very low density lipoprotein-cholesterol levels, as well as glucose level. The level of lipid profile alteration in Triton intoxication with renal ischemia reperfusion was significantly (p<0.0001) high compared to the Triton intoxication group only.There was significant (p<0.0001) correction of alterations inkidney (creatinine, urea and uric acid) and heart (creatinine kinase, coronary risk index and atherogenic index) function markers in the serum following post treatment with AMFJ.Moreover, results of the in vivo antioxidant assays revealed that AMFJ significantly (p<0.0001) reversed the redox imbalance caused by Triton intoxication on the various organs as adjudged by its effects on malondialdehyde and reduced glutathione concentrations as well as superoxide dismutase and lactate dehydrogenase activities. These results were consistent with the histopathological findings which showed amelioration of histoarchitectural abnormalities in the AMFJ treated group towards the control and sham state .In conclusion,this study demonstrated the hypolipidemic and protective effects of AMFJ against Triton-induced hyperlipidemia, and associated renal dysfunction. Renal damage causes increase in hyperlipidemia but the fruit juice ameliorates it effects compared to the Triton-induced with renal ischemia group, oxidative damage and atherogenic tendency in rats thereby lending credence to the juice as a potential therapeutic agent in the management of hyperlipidemia and related dysfunctions such as renal injury and damage. en_US
